We routinely currently work with MA104 cells in our laboratory. Recently we have been experiencing total cell death after about 3 passages after starting the cells up, irrespective of passage number. We have tested for mycoplasma contamination, which was negative and have change all consumables and have tested different trypsin concentrations. So far none of these changes have had any effect and the cells continue to die after three passages.

Is there something we are missing? Would you advise obtaining a fresh culture from the ATCC?
