I am trying to do some lentiviral infections of melanoma cells, the 293 FT cells always express the protein expression pretty well, but not the target cells. I used the same backbone plasmid having other gene as a positive control and get pretty good expression. My plasmid doesn't have GFP, so I check always by western blot only. With my positive control showing above 80% green cells, I know my packaging plasmids are working good (using second generation packaging, plasmid is FG12 backbone). My protein of interest is a Protease, one of my hypothesis is might be protein is itself killing the cells, where LV is infecting, is this a possibility? Although 293 ft are surviving and showing good expression. Is there any way to check, if the protein is killing the cells? One
