use methanol/water 75:25 pH=4 for extraction, and by loading the extraction in silica column, you can fractionate the extract by a fraction collector. Spot the fractionated extract of the tubes on TLC plate., and spray it by anisaldehyd spry. you can find whether you have good separation or not.
however, finding which solvent is the best in silica column is your job. in my case, after several times, I found what the best is for mine.
next step: you have to charachtrize the components inside every tube by LC-MS, NMR or X-ray.
this is the simplified procedure that I explained to you.

The first step is to determine the type of compound you want to isolate from the plant, for example, the isolation of alkaloids using several solvents such as methanol chloroform ethyl estate and if you want to separate one compound using chromatography technique
Each step in chemistry is followed by a bioassay
So you cannot predict in the begining the family of compounds. We will isolate the active compounds and determine the structure.
Since you know the type of compound to be extracted, the extraction method to use depends on the polarity of the compound desired. (The general trend is to extract with increasingly polar solvent). The crude fractionations from each extract must be tested for bioactivity. Use chromatographic separation methods (Column, Prep TLC, HPLC.etc.) and test the bioactivity of each fractions until you get your desired compound. The literature will help a lot as the phytochemistry of specific species in that genus must have been done before.
It depends on which type of metabolites you needed for your study.most of flavonoids and phenolic content of plants showed anticancer potential..
