I have questions regarding limiting dilution on primary MEF. I read previous discussions (https://www.researchgate.net/post/Is_dilution_cloning_in_immortalized_mouse_embryonic_fibroblast_MEF_cell_lines_obtained_by_serial_passaging_feasible) . Limiting dilution seems ok with immortalized MEF. However, I have lentivirally-transduced primary MEFs that have passage no. is 5-6.
I would like to knockdown genes via lentiviral transduction in primary MEF and do limiting dilution for selection and pick out single colony. Does anyone perform this procedure before? Is it successful?
