The staining with Giemsa suffers interference from the pH of the dye solution. If I remember well, more acidic solutions make the nucleus get a reddish coloration. On the other hand, more alkaline solutions make the nucleus bluish/violet.
ELISA is an indirect method of diagnosis is it easy to to be used and replace direct microscopic examination Which may be need well trained experienced staff !!
