We recently purchased a canabinoid receptor KO mouse strain to use for our experiments. Although genotyping so far showed that the mice were true KOs, when we isolated cells from the BM of these mice, they responded well to treatment with selective agonists for this receptor.

We then ran a qPCR and discovered mRNA transcription in all tissues the receptor is expressed in WT mice; spleen, brain and lungs.

So, the facts are that the cassette is indeed disrupting the gene (that is confirmed by genotyping), but mRNA and protein is produced (confirmed by qPCR and the functional test with primary cells). However, there could also be another explanation where another receptor could signal for the response of primary cells ex vivo. Therefore, the question is: can you suggest any possible way to find out whether WT, functional protein is expressed?
