We used to have a Cary 300 spectrophotometer at the lab, to perform mitochondrial complex analysis, using the kinetics mode.

We no longer have the instrument, and I need to know the following:

did the instrument substract the reading of the blank automatically for every point of the sample reading, along the whole experiment? Or did it just zeroed itself just once, prior the beginning of the analysis?

I can see that the instrument plotted the curve of the blank, but it was not integrated between the same points than the samples, to give the slope.

And I couldn't find a clear explanation of it while reading the user's manual.

Do someone has experience with this?
