Hi, I've done a karyotype method by using nocodazole 0.1 ug/ml. The method goes with obtaining liver and culture it in rpmi 1640 media overnight. Then, I added nocodazole and incubate in room temperature, 0.56% KCL, Carnoy's fixative and used Giemsa Stain. The normal procedure. But all I got was this picture shown here.
1. I couldn't identify this round thing here. Do any of you know what these are?
2. What is the problem here to cause me to get these result instead of the normal or supposed chromosome picture?
Hello! On this picture shows mitotic cells. But I think that you didn't get hipotension, there are not flights. Try to increase time of exposition of the hipotension.
