I am wondering if I can do the manipulation of the microorganisms on the bench and then incubate them anaerobically on pre-reduced media in jars, including an anaerobic generator bag.
Will a brief contact with oxygen affect bacterial growth?
And will the contact with oxygen oxidize the reduced solid media in the petri dishes, during the inoculation?
little amount of oxygen is toxic for the obligate anaerobes. They will be died due to contact of oxygen. Although, I think oxygen can oxidized the reduced media.
Strict anaerobic organisms don't require even little exposure to oxygen, oxidation of the reduced medium is possible. Facultative anaerobic will be your end Produ. Use the anaerobic chamber to get the strict anaerobic organisms first, multiply (grow) in suitable anaerobic medium before manipulation.

It depends on species with you are working with.
I am working with C. difficile (strict anaerobe) and the literature says that 30 minutes on oxygens kills vegetative cells. But during my expretiments I found out that they are much better oxygen fighters. Simply, try to be fast as you can to make the oxygen exposure as short as possible.
Regarding culture media. I use commercial ones (selective for C. diff. and non-selective Schaedler). They are packed at the same condition (plastic bag) as media for anaerobic culture. When I unwrap them I do it in room temperature as well as atmosphere. Also, I prepare Wilkins chalgren agar by myself and again I do it in laboratory.
