Strange 🤔 but I came across a very weird issue. Actually, I have been using a destination vector (pMDC85) 2x35 GFP since quite while and got no issue for transient gene expression in tobacco. But suddenly, I got stuck with two clone in the same vector for not getting any GFP expression transiently as I checked with sequencing result from promoter-gene-gfp-ter and seems everything is fine and in frame.
For the information I also used p19 along with the plasmid but got nothing.
Again, same vector work just fine for other gene. So, I am blank with the outcome as sequencing of the whole cassette is fine even done twice.
Any suggestion would be highly appreciated🙏
