Dear Sir:

Mixed coccidial species infections in poultry is very common. To the best of my knowledge, there may be No or the ones present are not useful methods for separation of different coccidial oocysts from a mixed infection. For experimental infections you need to have viable oocysts. Anay interference that may result in dead oocysts is not at all recommended. The only practical way- as far as I know- is to look for single infections within the tested population. You need to incubated these oocysts in 2 to 2.5% potassium dichromate solution at room temperature (26-30C) and give them the time needed for reaching full sporulation. Fully sporulated oocyst contain four sporocysts each with two sporozoites (eight sporozoites in every oocyst).

You could isolate a single oocyst (after sporulation) by micromanipulation and then try to build up the number of parasites for subsequent experimental work. See:
