i am differentiating my podocytes cells which require temperature incubation at 37 degrees for 14 days. i notice that a certain portions of the plate the cells look different. the first attached picture are cells that i would describe as morphology of the undifferentiated but in the 2nd attached picture the cells look like cobwebs. are these bacteria contamination/cells not healthy or are the cells undergoing differentiation?

It doesn't seem to be bacterial contamination here. I don't know this cell type, but I would bet cells are differentiating.

No contamination here, I feel it might be cell necrosis due to toxicity. check your differentiation media concentration.
I don't see bacterial contamination, but some cells are not so nice with retractile fibers (necrosis or apoptosis?). Probably, you have to change your media more often.
There is no sign of contamination. I am not sure whether the protrusions I see are signs of cell suffering or differentiation

hi thanks for all your valuable advise. 3 days after this question, i again see weird shapes in my culture. I will be happy if anyone is willing to take a look at the images attached in the question link above.

The cells look healthy and the picture is quite clean and clear. However, bacterial contaminations change the media color, the cells look turbid which makes very difficult for a clear image. The cells are in stress definitely which can be seen from the bright dying cells that might be floating around. Cannot say whether they are undergoing differentiation or not but certainly, morphology seems to be changed which is a primary indication of differentiation. You need to look into other factors as well that support differentiation. I would suggest you change the media with a mild wash with RT PBS or same spent media.
