We have been having a cell culture infection issue with our mESC (see photo attached).
These tiny moving particles can be seen between mESC colonies 2-3 days after seeding them and seem to have a small effect on cell proliferation, without altering the pH.
I find that this maybe a mycoplasma infection but is it possible that is it not?

to my knowledge the mycoplasma is soo small to be not detectable on microscopy but you can figure that out, by performing the mycoplasma detection by PCR.
It is a simple PCR test that you can perform on the cell surnatant is less that 3 hour.

Agreed, these are possibly too large to be mycoplasma; yet, these particles in between my colonies are of bacterial nature, no?
i agreed with the answer of Manuele Martinelli, but then try to use few antibiotics (Minocycline, Tetracycline, Ciprofloxacin, Enrofloxacin and Sparfloxacin) to eliminate the mycoplasma in your cell lines and then if you still see such colonies then sent it for Mycoplasma- PCR or DNA staining or ELISA etc. which will help you to find the contamination with mycoplasma as its not possible to see or confirm with microscopic images ...
