Do you have a sequence for your protein of interest? What tissue are you trying to stain? When you say, "there is no reliable antibody", do you mean there are no antibodies to your protein or that there are some antibodies available but they do not conform to your staining protocols? There is a big difference between the two.

Thanks for the reply, we are staining brain sections and we do have few antibodies but not very good results

Have you tried to optimise the antibody at different concentrations when doing IF and also optimise the blocking and secondary antibody concentrations

Is it a nuclear or surface protein? Do you have a KO as control? Can you do RNA FISH instead of staining?

Thanks everyone we have tried everything, its a nuclear protein and yeah haven't attempted FISH so far.
