Hi, you can do it following different ways:

1. WB, using ratio between mitochondrial protein and loading control (TOM20/Actin i.e)

2. Fluorescence microscopy. stain mitochondria and relate fluorescence intensity to cell area.

3. Flow cytometry: using Mitotracker Green you can achieve the mitochondrial content. This dye is independent of membrane potential so, it is not affected by mitochondrial dysfunction.
