Dear Hafiz,

for fungal DNA isolation all you need is enough biomass. You can be successful by either growing a liquid culture, collecting the dry biomass and grinding it for extraction or growing on solid medium and collect from there.

For most fungi growing on PDA and collection after one week should work. If you have any more questions just send me a message! Happy to help

i would like to know more that :

1: if some fungi grow slower ( took about 1 month for petri dish completion) and some faster (1 week for petri dish completion) so what should do in that case for sample collection for DNA extraction?

2: Can use more then 1 plug of same isolate in one petri dish for getting more quick material for DNA extraction?

3: Isolate sample collection for DNA extraction : one isolate per petri dish colony should collect in one 2 ml and put in liquid nitrogen and then grind and use CTAB or any kit ,,,,is ok? how will know that the sample is 30 mg for DNA extraction??

4: Spore calculation through haemocytometers procedure: can you please share a general formula for spore calculation ? as i have read many articles and questions but mostly labs used there own method and formula, ( pores/ml = (n) x 10^4, can explain this what is 10^4, ?

5: can you please share, that How can make the suspension solution for spore calculation through haemocytometers?

6: If i want to do inoculation of that isolates on fruit with different connections then how can make different concentrations?
