I am performing co-fermentation experiment with glucose and xylose and to determine the residual sugar concentration I generally prefer HPLC method but HPLC is not in work for a few days. Can anyone suggest me a way to estimate glucose and xylose concentration separately because I want to see how much xylose has been consumed ?
By polarimetry measurement (measures of optical rotation) you can differentiate glucose-xylose, because the glucose has a specific rotation of 52.0 +/- 0.5 and Xylose 19.0 +/- 0.5 . Well HPLC methods are best if you want good results, you can also try with GC as well (by derivatization of sugars using TMS reagent).
You can try with Phaloroglucinol assay for Xylose, a colorimetry method (I am attaching the protocol file). For glucose estimation you can look for glucose oxidase methods (an enzymatic method).
Also try with anthrone assay if looking for mixture of sugars.

A alternative way will be mesuare the reducing sugars by DNS method (for exemple), and the glucose by enzymatic kit (GOD-POD). By difference you will know the quantity of xylose.
