Hi, I have a question about the binding affinity of Fc receptors including FcRn.
It is known that Fc-silent antibody does not bind to Fc gamma receptor. However, I wonder if it does not bind to FcRn or if the binding affinity is only slightly decreased.

Fc-silent means generally with blocked Fc effector functions like ADCC. The key aim of engineering there is blockade of effector Fc functions which can induce an immune response. The goal is not to affect or make a shorter half-life. Consequently as was concluded in the paper below:
"They show no detectable binding to Fcγ receptors or to C1q, are inactive in functional cell-based assays and do not elicit inflammatory cytokine responses. Meanwhile, binding to FcRn, manufacturability, stability, and potential for immunogenicity are unaffected."
