am running sds page and now based on the molecular weight of resulted bands i need to identify the possible enzymes or protein present in my sample.
So, is there any database, by which i can search the possible enzymes, by entering the molecular weight of resulted bands ?
The subunit molecular weight of a protein on a gel cannot be used as a method of identification. If you have enzyme activity, you should measure those activities using specific substrates.
To go from bands on gels directly to identification, you would need to be sure that all bands are completely resolved in the gel and then do sequencing (the sequence also has to be known and in a database).
If you strongly suspect certain proteins, western blotting with highly specific antibodies may aid identification, but sequencing is more rigorous.
If the target is identified in the database, excising each band from the gel and subjecting them separately to an in-gel digestion procedure is the appropriate workflow for bottom-up proteomic identification. De-novo sequencing can be performed if the enzyme/protein is not available in any database and for this, you need to extract intact protein from the gel by means of electroelution or using alternative extraction techniques.
