I'm doing characterization of optimum temperature for bromelain enzyme from Bogor pineapple by measure the activity of the enzyme. I used casein 1% as substrate dissolved in universal buffer pH 8. The result showed that activity of bromelain enzyme more stable at high temperature, but labile at room temperature.
The enzyme are fully active at their optimum temperatures. The enzyme under study may have such a conformation that at low teperatures it does not assume its optimum active conformation and looses its activity. As the temperature increases, the enzyme structure may assume a conformation that activates the enzyme and stabilizes it.
