I want to compare the amount of an enzyme's expression (laccase for example) or presence in the tissue as a result of the experimental treatment. Is there a protein similar to an antibody that might bind with the enzyme and can be filtered then quantified by spectroscopy? or perhaps by comparison of PCR bands. I am interested to hear any other thoughts or suggestions as well. Thanks very much.
The activity of an enzyme can often be measured in a complex mixture if there is a specific activity assay for that protein. You will require a substrate and a means of detecting product formation or substrate consumption. You can set this assay up yourself based on information in the literature or use a commercial kit. The amount of a protein in a complex sample can also be measured immunologically using an ELISA. This requires at least one antibody or antiserum that specifically recognizes the protein of interest. If the protein is available in purified form, that can be used as a standard for quantitation. It can also be used as the antigen to raise the antiserum. If there are no commercial ELISA kits for your protein, you may have to work out the method yourself.
