Is there an acid treatment that allows removal of plasma membrane protein ???

I would like to determine the relative percentage of a protein expressed at the cell surface as well as intracellularly and I remember that there is an acid wash treatment able to destroy membrane protein without affecting too much the plasma membrane. Does anybody know the details of the protocol ??

Hi Dario,

so far I know, there is the possibility to remove "peripheral membrane proteins" or membrane-associated proteins relative selectively with either alkaline or acidic treatment. Mostly with bathing the membranes on ice in high pH-buffer (pH8-12) or low pH-buffer (pH3-5). But this treatment is not sufficient to remove integral membrane proteins.
