I am using the hind limb ischemia model in NOD/SCID mice and would like to measure changes in metabolism to the localised tissue both in a control mouse and mouse with recovered hind limb perfusion following hematopoietic progenitor cell transplantation.
You are mentioning immunohistochemistry. As far as my experience is, there are lipid stains to stain the total lipid and there are carbohydrate stains to stain the total carbohydrates in the muscle. The alternative I can think of is to study the metabolism in the frozen muscle sections is to stain for vital dyes in presence and absence of sodium azide (inhibitor of glucose metabolism). Unless you have a specific lipid in that case use the tracer (C14 labeled glucose or lipid) to study the metabolism
