I am working with sea cucumbers, and i am looking for a method for isolating plasma membrane lipids from a tissue.
There is quite a bit of literature on analyzing plasma membrane proteome of cultured cells, but I havent managed to find a concise method for lipids and complex animal tissue.
Do you have a specific cell type within the sea cucumber you want to look at? If so, I would start by isolating the cells of interest followed by a Bligh and Dyer lipid extraction on the crude, isolated cells. Plasma membrane lipid extraction has been done many times on many different cell types.
Our lab has previously isolated and extracted the cardiac (as well as hepatic) mitochondrial phospholipidome from mice and used the purified lipid extracts for analyzing various endpoints of mitochondrial membrane structure-function.
