How can we determine the amount of DNA in the tissue without using a commercial assay kit? We are looking for a simple procedure.
DNA after extraction? You can use spectrophotometer without any kit.
As Dr Konstantin A. Blagodatskikh suggested, you can use spectrophotometer. You must first extract the DNA from tissue samples.
Thank you so much for your valuable answers. We will quantify the amount of DNA by a Nanodrop spectrometer, but we need to find a simple DNA extraction method.

It depends on the source of your DNA. If it comes from bacteria and you want to isolate plasmid, there are "house made" protocols to isolate the DNA without any kit. Same for DNA from plants or for animal tissues. If you are more precise, I can tell you exactly what we use, kit-free
