Researchers have often grown yeast on non-fermentable carbon sources such as glycerol, particularly when studying processes such as mitochondrial function. The standard media would be :

Per liter:

Yeast extract, 10g

Bactopeptone, 20g

Glycerol, 20ml

Add purified water to make the total volume 1 liter.

If you need to use a defined media, you can use Bacto Yeast Nitrogen Base which provides salts nutrients and nitrogen source.

Since this media is defined it might help in your analysis

you can use minimal media (-) C source and add glycerol as C source ...or use PDA .....for cultivation and C- testing

The most common amount of carbon sources (incl. glycerol) used in assimilation tests is 1%.
