My work in sequence analysis brings me to the development of novel assembly software, and therefore I have need of a set of reads sufficient to test that software. Reads that are ready for assembly is my quest. Can you supply?
The goal is to develop a novel genome sequence assembly software tool, one that yields complete genomes by chromosome. For that, I need real sequence data. As I approach this problem from a strictly computational point of view (since I know not the intimate details of attendant chemistry and results interpretation), the naive answer seems most appropriate - to wit, whatever NGS technology is suitable for the demonstration of the algorithm I've got in mind.
Respecting collaboration, yes, I am looking for such.
Please, note that you can also use simulation software like the one mentioned here https://www.biostars.org/p/2194/
The advantage of using simulation is that you know beforehand what the assembled sequence should be. But SRA has many sequences from organisms with good reference genomes, too.
