I am working on subcellular origin of puffer fish toxins and I need to detect the presence of Tetrodotoxin in crude tissue extracts of the fish through GC-MS. Is it mandatory to make derivatization using Trimethylsilyl compounds to my crude extracts to make tetrodotoxin volatile and make easier to analyze it?
Yes, you can run your sample on GC-MS and try to observe the chemical sketches than you proceed again either for crude/purified sample.
Depends on the analytical pathway set up on your GC-MS. If you use a column with a non polar phase (e.g.HP5 or HP1 phase or equivalent) you will need to derivitise it as it will interact with the phase too much due to tetrodotoxins high level of polar functional groups. A polar phase (HP-210 or HP-225 or equivalent) column should resolve it acceptably underivitised, though the volatility may still be an issue as its melting point is quite high (225°C). Increasing volatility will make it easier overall to analyse.
