ORAC is a powerful tool to determine the antioxidant capacity of a test solution. My experiences with rat/human/chicken/mice plasma has taught me that testing these at correct dilution of paramount importance. Make sure that you are estimating the antioxidant capacity in the linear range of the standard curve and you have rigorous controls for your assay. It is very easy to get false positives during ORAC due to high antioxidant capacity of serum proteins. Since you are just testing plasma, I reckon that ORAC hydrophilic test should be enough as I don't expect non-polar or lipophillic compounds to be roaming around in plasma.
Dear Mohammad Assi I hope you have to repeat not less than 5 times to get the result with plasma because it is very rich in protein and other metabolites.
If you want to get rid of the proteins, use protein filters and concentrate your sample. Lyophilise the residual water which will contain the hydrophilic compounds which are free and not protein bound. Use this to get an estimate of the anti-oxidant capacity. After all protein bound analytes are rarely active.
