Beta gal activity is considered as a hallmark of senescence. The enzyme activity is significantly amplified in senescent cells due to increased lysosomal content. Morphologically, senescent cells become flat, enlarged and vacuolized and sometimes appear with multiple or enlarged nuclei. Senescent cells also form cytoplasmic bridges that allow them to signal to the neighbouring cells via direct intercellular protein transfer. I feel they are showing senescence. In addition to cell morphology, the beta gal assay would help to confirm the results. Did you run a control
(without treatment)?
