we need to purify the pyrrolidone derivatives. When we checked the TLC, the compound Has Rf of 0.6 in acetone. But other solvents showed no movement of the spot from the base line. However, when I used acetone to pack the column we noticed white solution of the acetone collected.
Please mention any references that are using only acetone if possible. Kindly help us with your suggestions.
No, silica gel isn't soluble in acetone by itself. It isn't polar in more polar solvents such as methanol, and is slightly soluble in water (much less soluble in acidic water). Silica does dissolve in basic polar solutions, greater than pH 7.5.
See: Alexander, G.B.; Heston, W.M.; Iler, R.K. The Solubility of Amorphous Silica In Water J. Phys. Chem. 1954, 58(6), 453-455 for a discussion of silica solubility in methanol and water. The solubility was determined after allowing the silica to interact with the solvent for several months. As acetone is much less polar than silica, there would be correspondingly less opportunity for silica to dissolve.
Thank You Jack Silver. Most relevant answer I got so far. I performed the column chromatography by using 100% acetone. So far I got no silica in the collected solutions. But Initial packing of the column showed white residue in the solvent collected. I wonder what it might be?
