I used a mixture of Pluronic F127 and F127DA to form a hydrogel with cells encapsulated inside by crosslinking with UV. It seems that the cells show very poor attachment to the hydrogel, also less than 30% of the cells encapsulated were found to be viable overnight. The percentage of F127 used was quite high (over 20%). The photoinitiator used is about 0.1%w/v of Irgacure 2959 and UV exposure of about 5 mins at 1mW/sq cm which seems reasonable as per literature available. I am wondering if the material per se is toxic to the cells? i.e F127/DA
