We are working on different stress models targeting platelet aggregation. I would like to know whether platelet aggregation more in any of stress condition. Is there any condition that favors platelet aggregation before addition of agonists in Platelet rich plasma (PRP)? If anyone expert in using CHRONO-LOG® Model 700 Whole Blood/Optical Lumi-Aggregometer, please let me know if platelets been aggregated already, is this instrument give accurate results particularly in stressed platelets from animal models (stroke, diabetes, Alzheimer's and Parkinson's). Please help me out from this by providing suitable answers and articles. Thank you for your kind attention in my query.

We have tried in different types of stressed platelets in aggregometer but had no luck.If platelets are prone to aggregate in stress conditions, is platelet aggregating agonist concentration and platelet count in aggregometer is really matter to achieve adequate aggregation. Please suggest me how to proceed if platelets are not responding in stress condition.

Detecting preformed platelet aggregates with an aggregometer is complicated. Impedance bases on platelet aggregation on the wires, preformed aggregates can't be diffentiated from actual aggregation. Also, aggregometers assume the initial sample to be aggregate-free and uses this for calbration of the sample signal.
One way would to try spontaneous aggregation without using any agonist. Flow cytometry using e.g. CD41/CD62 as markers might be the preferential method using the platelet size distribution to differentiate between microparticles/microvesicles, platelets and aggregates. CD41-positive microparticles (or other platelet markers) are also a good marker for in vivo platelet activation, as they are formed during the activation process.
