I am trying to quantify BRDU/DCX positive cells, but have had bad luck with both of the antibodies working. BRDU will only work with HCl pretreatment. DCX will not work following HCl treatment, but in this case before HCl treatment it seems to work. Could you please tell me if I should pursue this order in antibody treatment? 2X SSC formamide treatment followed by NeuN primary and secondary over night, followed by DCX primary and secondary, followed by HCl pretreatment and BRDU primary and secondary.
Thank you for your help and your input.
Also, this is being done in P6 mice, so they might be staining differently.
In the image below the green is DCX, red is BRDU and blue is NeuN.
