Greetings to all researchers! Can anyone confirm weather it is true or not that cotyledonary node are not recommended to use for in vitro multiplication when true-to-type is the purpose of the work? and if yes, why?
Under circumstance cotnodes are and efficient system but there is the chace of ploidy variations probably for cytokinins effects on swuch high regenation rate tissues. http://link.springer.com/article/10.1007%2Fs11240-004-8833-5?LI=true
Based on the media composition if your media contain 2,4-D ,the chance of getting variation true but with less than 10% due to the plants derivative from callus .
Although axillary bud culture is regarded as an ideal method for the production of true to type plants but still variations may occur in plants derived from the axil of same cotyledonary node even without intervening callus phase. This might occur due to the stress produced through wounding etc.
