I am working on cell lines. I wish to know if its suitable to keep cell lysates at -20 degree celsius for a period of atleast one week, before performing enzyme analysis with the cell lysates.
It would in large part depend upon the enzyme in question. If it is known that the enzyme activity is stable at -20 degree C, it should be ok. If not, add 50% glycerol (sterile) and store at -80 degree C. In some assays, the presence of glycerol interferes with the enzyme activity.
I think less than a week time is ok to store cell lysates in -20, though it could affect the enzyme activity. With my experience I can say that you can still get enough enzyme activity if you use lysate within a week but that too depends upon the particular enzyme that you want to assay. Try to store your cell lysates in -80.
