I want to do golgicox staining , but a cryosection and vibratome facility is not available. Is it preferable to use a paraffin embedded tissue section?
I have not stained for Golgi using paraffin-embedded tissue, but I have used Cresyl violet stain in paraffin-embedded tissue (Hippocampus 1995 vol 5, pp 1050-9631). I recommend contacting FD NeuroTechnologies to inquire whether the Golgi stain would work? An alternative is to work with a collaborator (or pay a company) to process the the tissue for you.
I think you can follow the standard procedure for Golgi staining and at the end prepare paraffin embedded blocks of already-Golgi stained samples. There should not be any problem with your sections. Check the following publication please:
