Is it possible to stain histological slides from larger tissue sections and without dissociating the cells, with Acridine Orange or Giemsa dyes aiming to verify micronucleus formation?
We intend to verify DNA damage caused by environmental factors in a specific tissue, but it would be much more helpful to observe this in a whole tissue section. Did anyone ever try this idea?

Thank you my friend. I was searching through my papers library and I already have this review. I'll take a better look because I thought all the mentioned studies used dissociated cells. We'll give it a shot with PAS staining too.

Histological sectioning reveals details about the tissues of the organ analyzed rather than the cellular components.
I will read the suggested paper in Mutat. Res. again myself and give a final suggestion in this regards
One of the histiological variables is nuclei count and their morphological measures in any tissue , just add a new one , the MN count, but check the dinamic of the tissue considering the begining of the mutagen exposure,
- tissue dissociation if less expensive with less time and you dont need an histology lab, the inclusion of water in the citoplasm with hypotonic treatment of cells is another advantage let more area to score,
but these features dont mean that you can use histological section to count MN.
