I'd like to stain bacterial cells to see all of the cells' nucleiods along with which cells have disrupted membranes. Can DAPI and PI be used in the same cells? I know their emissions are very different, but what I mean is if one dye would displace the other because it binds the DNA stronger? Has anyone done this? A link to a paper would be great! Thanks

I think you can use both of them at the same time. Because DAPI is a DNA minor groove agent and PI is an intercalator. This means they are going to bind different parts of the same DNA.

I've used PI in vivo then followed with DAPI and haven't had a problem. Granted I have not fully quantified the % of overlap between the two signals.
This paper can help you. I did probes to evaluate biofilm formation and I used DAPI and PI at the same time.
