I am at the certain stage where it would be better to know the sequence of my RNA of interest. I have the question about sequencing of the PAGE purified RNA. I am thinking to prepare the cDNA of the specific PAGE purified RNA (from single band). I am having right primer. Will I be able to know the exact 5'end of the RNA.
I would be highly thankful if someone would provide me the possibility and suggestions about it.

Hi there,
For RNA sequencing you have to go to the cDNA first in order to run the sequence on a DNA Template. If for instance the whole cDNA is cloned into a plasmid then full sequence would be accessible using plasmid specific primer extension.
