I have access to NMR instrument which is used only to assay samples in the liquid state (dissolved form). Kindly suggest any protocol. I have access to a few literatures which says a mix of ionic liquid and DMSO can dissolve biomass. But I am not sure if it will be good for assay.

Perhaps I'm not understanding you assay, but I suspect that doing assay by NMR will be difficult.
- Plant extracts (I assume this is what you mean by "biomass") are initially a mixture of many things. It is difficult to get a PMR or CMR from that. You might be detecting on another element (hypothetically, flourine) assuming the compound of interest contains this element. Perhaps you have an isotopically labeled compound, where the isotope is NMR active, that the plant incorporates into the product? Maybe 13C? Otherwise using natural abundance isotopes, it would be difficult to resolve a particular compound from the mass of signals, and you would have to know the spectrum of that compound.
- You would need a known compound (or class of compounds) to know which proton or carbon peaks to look for, and those peaks would need to be resolved from the signals from the rest of the extract, assuming no NMR active isotopic enrichment.
- You would want to use deuterated DMSO
If you are using isotopic enrichment, also consider mass spectroscopy as well; with a known compound, one can do mas directed fractionation.

I am sorry for being little vague by calling 'plant Biomass'.
My purpose is to analyze the dry plant biomass for cellulose hemicellulose and lignin, (to study degree of delignification treatment)
As you have suggested deuterated DMSO, will this work to dissolve powdered dry plant biomass so as to make is liquid NMR compatible?

We understand the question better, but I'm sorry, I've little knowledge of how these compounds dissolve in DMSO.
