In vitro, DNA ligation can be tested by a variety of methods, the most common being agarose gel electrophoresis in the presence of urea. To test it in vivo, you would have to take a sample, extract the DNA, and perform the test. To recognize the specific piece of DNA, you would have to label it in some way, such as with radioactivity, biotin, or a fluorescent dye.
