How can I measure MMP-1 protein activity or protein level from liver tissue?

Instead of checking only the mRNA level, I want to check the active protein level of MMP-1 in Liver tissue from mice. How can I do that?

Hi Garyfallia, you can isolate lipid droplets (LDs) from a frozen tissue and determine proteins of LDs fraction by Western blotting. Isolated LDs is not a good method to check their morphology. You can analyze LDs morphology by frozen section with Oil Red O staining or paraffin section with H&E staining. Good Luck^^ Luke

Hi Luke! Sorry for taking me so long to thank you. Well I have tried the Lipid droplet isolation kit of CELL BIOLABS (https://www.cellbiolabs.com/lipid-droplet-isolation-kit?gclid=EAIaIQobChMI8p3ty4Ga1QIVAhbTCh2Qtw0rEAAYASAAEgItFvD_BwE), but I am not quite happy with the amount of extracted Lipid droplets of adipose tissue obtained nor the expression of PLIN1 that was really low. Any suggestions? Any protocol that you have in mind? Thanks a lot anyway!
