I doubt why you dont have same types of papers.

Dont use raw curcumin but use curcumin and nanocurcumin for both.

Procedure for MTT assay are same (i.e same procedure with equivalent amount of same constituent).

You may check how many times activity are enhanced.

in general, in MTT target compound solution in DMSO was added to cell line wells. if we prepared the solution of curcumin and curcumin nanoparticles then nanoparticles get soluble in DMSO then they loss particles nature then there is no difference between curcumin and nanocurcumin.  there is no difference in solution state whether it is curcumin or nanocurcumin. then how can we say nanocurcumin has better/enhance anticancer activity than curcumin.

no need to disolve in DMSO if NPs are soluble in PBS 7

If not soluble in PBSPBS 7 , if soluble then nanoparticles loss the structur ? If loss how is comparable ?

Solubility is always a major problem… If the conformation of the nanoparticle is not preserved under the conditions of the assay (as compared to that of curcumin), the assay does not have any significance, excepted if it is demonstrated that the change in conformation allows to uncover the active site of the particle… This is not a trivial problem, and the methods that can be used to demonstrate this possibility are not simple tu use

for anti cancer activity we made an assay that is close to in vivo conditions if particles r soluble i PBS but loss conformation u should change ur protocol for preparing NPs not MTT assay as drug or drug NPs have to face PBS (blood in body) 

ur explanation concluding that it is not possible. is it right ? i too don't want to change the MTT assay etall. however, thank you for valuable suggestion.

yeah. it means we have to prepared a nano particles in such a way that deliver the drug in the form of nano particles and carry it into the cell. eg, loading the drug in different polymers, material, binding to different carriers. which make drug to reach cancer cell then released the drug after that drug affectively shows its activity compare to raw drug.
