I will try to express proteins in HEK-293T cells. I had previous experience in another cell line where I used co-transfection for selecting transfected cells. I had observed lower levels of protein expressed as the experiment went on. Totally understandable as there is no pressure to keep the encoding plasmid for the protein.
With this, I would like to avoid my previous problem and would like to transfect the HEK-293T cells with a single plasmid that expresses the protein and the selection agent. I have been looking for it in papers but no selection agents were described.

I wonder what is Your target. IMHO this vector may provide efficient transient expression, that would be prolonged a lot by SV40 Ori in episomal fashion in HEK293T Cells (but not HEK293 !!! ).
There is no mammalian selection antibiotic resistance in the plasmid so You may play with transfection efficacy only.
