I isolated a strain of endophytic Fusarium sp. from hibiscus leaves, but after growing the particular Fusarium sp. in a broth containing soy bean, the cultured colony differs from the one I first isolated when I sub cultured it back on PDA.
Could it be that the Fusarium sp. colony differs due to its "special form" (forma specialis /f.sp.)?
It is not usual to have so much morphological difference between original colony and the subcultures, even in Fusarium. Your culture is probably contaminated with one of those contaminants. You are usually not able to distinguish between form specials of Fusarium species. Best wishes.
I was surprised too, but I can confirm that it is not contaminants as the culture was pure from when I isolated it (used streak method on plate) before I sub cultured it inside the broth. I also did microscopy check for both colonies and they have almost identical conidia and hyphae. I have no explanation for this occurrence :(
