The predicted size for protein of interest (POI) is 40kD, however, I did not see a consistent band there while an obvious band was detected at size ~70kD in KD and KO experiments of POI. Is the 70kD band my POI?

do you know if it predicted to be highy glicosilated?
Since the glicosilation in certain case may increase a lot the MW of the protein respect the one expected just fro the AA sequence

Do you mean the glycosylation? I got some possible explanation for this from references which also mentioned glycosylation. I will verify this.
