Dear all,
I do have human embryonic stem cells. I differentiate them into NPCs. However, when I perform qPCR for various genes, I also detect cT values for tuj1 in stemcels. In neurons, the expression is increased 6 folds.
I also check for nestin, neurod1, synapsin and many other factors. They all fits together and indicate that my differentiation process works fine. I am not sure why TUJ1 is expressed in stemcells. Is 6 fold changes enough? Or do you think that my Tuj1 data is wrong?
Does anybody detect Tuj1 expression in stemcells before?

iPSC line pluripotency markers NANOG, OCT4, SSEA4, Lin28, DPPA4, REX1, TRA-1-60, Sox2, Klf4 and TRA-1-81. Neural progenitor cells (NPCs) markers are NESTIN, PAX6, and SOXs. When the NPC differentiate into mature neurons then it will have higher expression of TUJ1 and MAP2. 6 fold expression of TUJ1 in stem cell is really high.

I dont have 6 fold of expression in stem cells. I have 6 fold of increase in TUJ1 expression between IPSCs and mature neurons. I am was not sure if this increase is enough. Because normally I should observe "0" expression of TUJ1 in IPSCs. but I even see an amplified PCR band on agarose gel.

Did you ever figure this out, Enes? I am seeing TUJ1 in undifferentiated canine iPSC, by innmunocitofluorescence.
