Is it necessary to Peel off the volute if I want to extract DNA from cochlea?

The volute of cochlea is too strong to separate from the inner ear.

I am not exactly sure what you mean by 'volute'. If you mean the surrounding bone, then yes you are right, it is difficult to separate the Ductus Cochlearis from the bone. Unfortunately, to extract DNA from cochlear tissue, you really have to remove the bone. It needs a little bit of practice, and it is a relatively long dissection.
