I am adding a Halo-Tag to a protein that is already tagged at its N-terminal. I prefer my tags to be just at one side to leave the C-ter free. I plan to add the Halo between the two. Will this affect the activity of the Halo along with the fluorescent ligand binding? Or should I put it at the C-ter?
